NK Cell-Mediated Growth Inhibition of Tumor Target Cell in Normal Donors and Cancer Patients

Abstract
Using colorinmetric MTT assay NK cell-mediated growth inhibition of cell was measured in normal donors(n=36) and cancer patients(n=65) to replace the conventional radioactive base techniques for cell killing, In the first group the effector cells
of
normal donors(12 persons) were mixed with the K562 target cells(1¡¿10E3/well) at four effector/target cell(E/T) ratios ranging from 40:1, 20:1, 10:1, and 5:1 in an assay volume of 0.1 ml. In the second group the effector cells of normal donors(36
persons) and cancer patients(65 persons) were mixed with the same target cells as the first group at 40:1 effector/target cell ratio. After the optical density was measured at a wavelength of 620 nm the percentage-specific growth of the control
was
calculated. At the low effector cell concentration(5:1) of the first experimental group the value was over 100 percent. The values for normal and cancer groups at 40:1 E/T ratio in the second experimental group were 34.5¡¾16.3 and 71.9¡¾34.7
percents(p<0.005). These results showed that the colorimetric MTT assay system for NK activity had a probability to be used as an immune parameter.
